CKD
Adriamycin-induced nephropathy
Podocyte injury model of focal segmental glomerulosclerosis with heavy, sustained proteinuria.
Background and rationale
Adriamycin (doxorubicin) directly injures podocytes, producing nephrotic-range proteinuria followed by focal segmental glomerulosclerosis and tubulointerstitial fibrosis. It is the standard non-immune model for testing podocyte-protective and antifibrotic therapies.
Typical study design
- Single or split intravenous dose adjusted to strain susceptibility
- Stratified randomisation on week-1 albuminuria
- Serial urine collection to follow proteinuria kinetics
- Optional unilateral nephrectomy to accelerate progression
Readouts
- Urinary albumin-creatinine ratio and total protein
- Plasma creatinine, BUN, cystatin C, albumin and cholesterol
- PAS and Sirius red histology with AI-assisted glomerulosclerosis and fibrosis scoring
- Podocyte marker immunofluorescence and podocyte counting
- Electron microscopy and Magnephy super-resolved 3D imaging of foot processes
- Single nuclei RNA sequencing of glomerular cell populations
